An Assessment of the Technicon H-2 automated Blood Cell Counter

Abstract
Background:
The Technicon H-2 system, a new automated blood cell analyzer, introduced into our laboratory and so we evaluated the performance of the H-2. The H-2 performs CBC and WBC differential counts based on light scattering and cytochemistry.
Methods:
We estimated our laboratory's reference values of CBC and WBC differential count. CBC on the H-2 was compared with the STKS. And differential counts with H-2 was compared with method of manual and STKS. We evaluated the relative percentage of
carryover, linearity from diluted blood, and stability at room temperature and 4¡É.
Results:
the correlation coefficients of CBC between H-2 and STKS were 0.99, 0.98, 0.99, 0.97, 078, 0.86, 0.34, 0.80, 0.34, 0.80, 0.98, 0.61, 0.14 and 0.94 for WBC, RBC, Hb, Hct, MCV, MCH, MCHC, RDW, Plt, MPV, PDW and PCT, respectively. The
correlation
coefficients of WBC differential count between H-2 to STKS and (/) H-2 to manual were 0.96/0.94, 0.95/0.93, 0.62/0.48, 0.98/0.92 and 0.00/0.12 for neutrophil, lymphocyte, monocyte, eosinophil land basophil, respectively. The carry-over for WBC,
RBC, was
less than 1%. Linearity for WBC, RBC, Hb and Plt with samples of high values was good.
On serial dilutions, values f WBC and Plt were also good precision. The stability of WBC, RBC, Hb, MCV, neutrophil and lymphocyte were not significant changes between room temperature and 4¡É, except Plt.
Conclusions:
In this study, H-2 showed good correlation coefficients for CBC and WBC differential count when compared with STKS and manual method. And routine utilization of H-2 might have great impact on current large volume clinical hematology
laboratory
practice.
